    Extraction optimization and characterization of polysaccharide antioxidants from Pinellia ternata (Thunb) Breit rhizome

    Purpose: To investigate the optimum extraction conditions of polysaccharides from Pinellia Rhizoma (PRP) and their antioxidant activities.Methods: Response surface methodology (RSM) was applied to optimize the water extraction conditions of PRP by Box-Benhnken design (BBD). A high performance liquid chromatography (HPLC) method was performed for determining the monosaccharide composition, while a high performance gel permeation chromatography (HPGPC) method was established for determining the molecular weight distribution of PRP. In addition, scavenging DPPH and superoxide anion radical scavenging assays were used to evaluate the antioxidant activities of PRP.Results: As a result, the optimum extraction conditions of PRP were as follows: extraction time, 103 min; solid-liquid ratio, 24 g/mL; and extraction no., 3. Under these conditions, the maximum extraction yield of PRP was 2.47 %, which matched the optimum value (2.55 %) predicted by RSM. The results of HPLC analysis suggest that the monosaccharide composition of PRP was mannose (Man), galactose acid (Gal acid), galactose (Gal), glucose (Glc), and arabinose (Ara) in molar ratios of 5.76:2.20:2.64:4.57:1, respectively, and the molecular weight ranged from 13592 to 445065. Furthermore, PRP showed marked antioxidant activities with the highest DPPH free radical scavenging rate of 71.5 % and O2- free radical scavenging rate of 87.5 % at a concentration of 10 mg/mL.Conclusion: RSM is a rapid and effective statistical technique for optimizing conditions for PRP extraction.     Luminance Prediction of Paper Model Surface Based on Non-Contact Measurement

    The overall appearance perception is affected by luminance perception accuracy and efficiency mostly. The surface luminance prediction correlated with surface angle and surface tone value was performed by measuring and modeling the paper model surface luminance. First, we used a rotating bracket designed to facilitate to set the paper surface angle. Then, we set the surface angle from 5° to 85° at the interval of 5° using the designed rotating bracket. Additionally, the four primary color scales, cyan, magenta, yellow, and black, were printed and set at the designed angle. The angle-ware and tone-ware luminance was measured using spectroradiometer, CS-2000. Finally, we proposed and evaluated a mathematical model to reveal the relationship between luminance and surface angle and surface tone using the least squares method. The results indicated that the surface luminance of paper model could be predicted and obtained quickly and accurately for any surface angles and surface tone values by the proposed prediction model

    The next widespread bamboo flowering poses a massive risk to the giant panda

    The IUCN Red List has downgraded several species from “endangered” to “vulnerable” that still have largely unknown extinction risks. We consider one of those downgraded species, the giant panda, a bamboo specialist. Massive bamboo flowering could be a natural disaster for giant pandas. Using scenario analysis, we explored possible impacts of the next bamboo flowering in the Qinling and Minshan Mountains that are home to most giant pandas. Our results showed that the Qinling Mountains could experience large-scale bamboo flowering leading to a high risk of widespread food shortages for the giant pandas by 2020. The Minshan Mountains could similarly experience a large-scale bamboo flowering with a high risk for giant pandas between 2020 and 2030 without suitable alternative habitat in the surrounding areas. These scenarios highlight thus-far unforeseen dangers of conserving giant pandas in a fragmented habitat. We recommend advance measures to protect giant panda from severe population crashes when flowering happens. This study also suggests the need to anticipate and manage long-term risks to other downgraded species

    3D-Aware Object Goal Navigation via Simultaneous Exploration and Identification

    Object goal navigation (ObjectNav) in unseen environments is a fundamental task for Embodied AI. Agents in existing works learn ObjectNav policies based on 2D maps, scene graphs, or image sequences. Considering this task happens in 3D space, a 3D-aware agent can advance its ObjectNav capability via learning from fine-grained spatial information. However, leveraging 3D scene representation can be prohibitively unpractical for policy learning in this floor-level task, due to low sample efficiency and expensive computational cost. In this work, we propose a framework for the challenging 3D-aware ObjectNav based on two straightforward sub-policies. The two sub-polices, namely corner-guided exploration policy and category-aware identification policy, simultaneously perform by utilizing online fused 3D points as observation. Through extensive experiments, we show that this framework can dramatically improve the performance in ObjectNav through learning from 3D scene representation. Our framework achieves the best performance among all modular-based methods on the Matterport3D and Gibson datasets, while requiring (up to 30x) less computational cost for training.Comment: To appear in CVPR 202

    Toll-like receptor activation by helminths or helminth products to alleviate inflammatory bowel disease

    Helminth infection may modulate the expression of Toll like receptors (TLR) in dendritic cells (DCs) and modify the responsiveness of DCs to TLR ligands. This may regulate aberrant intestinal inflammation in humans with helminthes and may thus help alleviate inflammation associated with human inflammatory bowel disease (IBD). Epidemiological and experimental data provide further evidence that reducing helminth infections increases the incidence rate of such autoimmune diseases. Fine control of inflammation in the TLR pathway is highly desirable for effective host defense. Thus, the use of antagonists of TLR-signaling and agonists of their negative regulators from helminths or helminth products should be considered for the treatment of IBD

    Heterologous expression and characterization of a malathion-hydrolyzing carboxylesterase from a thermophilic bacterium, Alicyclobacillus tengchongensis

    A carboxylesterase gene from thermophilic bacterium, Alicyclobacillus tengchongensis, was cloned and expressed in Escherichia coli BL21 (DE3). The gene coded for a 513 amino acid protein with a calculated molecular mass of 57.82 kDa. The deduced amino acid sequence had structural features highly conserved among serine hydrolases, including Ser204, Glu325, and His415 as a catalytic triad, as well as type-B carboxylesterase serine active site (FGGDPENITIGGQSAG) and type-B carboxylesterase signature 2 (EDCLYLNIWTP). The purified enzyme exhibited optimum activity with ÎČ-naphthyl acetate at 60 °C and pH 7 as well as stability at 25 °C and pH 7. One unit of the enzyme hydrolyzed 5 mg malathion l(−1) by 50 % within 25 min and 89 % within 100 min. The enzyme strongly degraded malathion and has a potential use for the detoxification of malathion residues

    <p>Abstract</p> <p>Background</p> <p>It is widely considered that the multifunctional NS1 protein of influenza A viruses contributes significantly disease pathogenesis by modulating a number of virus and host-cell processes, but it is highly controversial whether this non-structural protein is a proapoptotic or antiapoptotic factor in infected cells.</p> <p>Results</p> <p>NS1 protein of influenza A/chicken/Jilin/2003 virus, a highly pathogenic H5N1 strain, could induce apoptosis in the carcinomic human alveolar basal epithelial cells (A549) by electron microscopic and flow cytometric analyses. NS1 protein-triggered apoptosis in A549 cells is via caspase-dependent pathway.</p> <p>Conclusions</p> <p>Influenza A virus NS1 protein serves as a strong inducer of apoptosis in infected human respiratory epithelial cells and plays a critical role in disease pathogenesis.</p
